Summary

Senior Research Associate with a background in nucleic acid chemistry. I hold a Master's in Pharmaceutical Sciences and have hands-on experience in combinatorial chemistry, DNA polymerase assays, and next-generation sequencing-based quality control. My work focuses mainly on improving experimental workflows, generating dependable data, and supporting integrated scientific teams. Over the past three years, I have been actively involved in small molecule drug discovery using DNA-encoded library (DEL) technology. I am currently seeking a full-time position.

Work History

Senior Research Associate

Terray Therapeutics
10.2023 - Current
  • Led a team of five research associates to perform combinatorial chemistry on solid-support beads, synthesizing over 5 million unique compounds per combinatorial library in a 96-well format. Built and QCed two libraries biweekly using chemistries including amide coupling, reductive amination, Suzuki coupling, Buchwald-Hartwig amination, and nickel sp2-sp3 cross coupling.
  • Optimized copper-catalyzed click chemistry for DNA tagging of DEL libraries, improving tagging efficiency from 82% to 97%. Served as primary contact for glovebox-based chemistry workflows.
  • Performed post-click QC of oligonucleotides using fluorescence-based hybridization assays to confirm DNA sequence identity and integrity to detect well-to-well cross-contamination.

Research Associate

Plexium
10.2022 - 10.2023
  • Developed the company wide SOP for high-throughput spectrophotometric DNA quantification by discovering the limit of detection for modified oligonucleotides. Improved efficiency, cutting processing time from 1 minute to 8 seconds per sample. Two hundred unique sequences were quantified weekly.
  • Automated solid-support primer extension reaction using the Integra Assist Plus, a benchtop liquid handler, optimizing reagent handling for volume variation, liquid viscosity, and concentration differences.
  • Optimized polymerase primer extension reaction on DNA-encoded beads, identifying a reduced reaction time that maintained QC standards. Cut process time from 1 hour reaction time to 30 minutes in a 96-well plate format.
  • Synthesized DNA onto solid support via primer extension reaction for DNA-encoded small molecule libraries. Performed QC utilizing qPCR and Illumina sequencing after each DNA extension and chemistry step to assess DNA damage and sequence identity.
  • Prepared and collected DNA samples for NGS sequencing. Fully responsible for DNA library preparation and operating the MiniSeq Illumina sequencer for the entire company. Sequencing was conducted twice weekly.
  • Executed Python scripts in Jupyter Notebook to analyze sequencing data, providing insights for the compound screening team to identify potential drug candidates. Facilitated the screening of over 80,000 compounds per library cycle.
  • Presented data to scientists, directors, and research associates on a biweekly basis, ensuring clear communication of DNA-encoded library QC results. Delivered high-quality data to the chemistry, biology, screening, and assay development teams to support research integrity and decision-making.

Grad Student

Dr.Andrej Lupták, UCI Department of Pharmaceutical Sciences
09.2020 - 09.2022
  • Managed an independent project involving DNA polymerase reaction kinetics where data was collected for the lab's NASA astrobiology grant.
  • Synthesized, prepared, and purified DNA oligonucleotides for Illumina sequencing via DNA primer design, PCR, and PAGE purification.
  • Performed initial computational data analysis of DNA sequencing data including quality control analysis of nucleotides, Bowtie2/BWA alignment to a reference genome, and nucleotide depth analysis on millions of aligned sequences for DNA pattern recognition.
  • Developed own script using Python for DNA nucleotide frequency quantification to search for differences in experimental vs control conditions for GB sized FASTA file format of Illumina sequencing data.
  • Oversaw and trained a team of 4 undergraduate research assistants for adherence to all lab procedures and protocols.

Grad Student

Dr. John Chaput, UCI Department of Pharmaceutical Sciences
06.2018 - 06.2020
  • Synthesized, purified, and identified various small molecule nucleoside analogs via organic synthesis, column chromatography/HPLC, and NMR.
  • Performed kilogram scale synthesis of nucleoside glycosyl donor for threose nucleic acid (TNA), an artificial nucleic acid derivative of DNA, optimizing production to 200 grams of material weekly.
  • Utilized the previously synthesized glycosyl donor to create all four nucleoside compounds (ATCG) for follow-up synthesis of both phosphoramidite and nucleotide triphosphates.
  • Maintained the DNA synthesizer for solid phase synthesis of artificial oligonucleotides. Utilized triphosphates for enzymatic synthesis.
  • Presented monthly research updates to a cross-functional team of both biology and chemistry students requiring fluid communication across both disciplines.

Volunteer Research Assistant

Dr. Amal Alachkar, UCI Department of Pharmacology
08.2017 - 05.2018
  • Monitored two distinct B6 mouse lines, including cage maintenance, breeding, and weaning. These mice were genetically modified with cre-recombinase and lox P sites for tissue specific receptor ablation.
  • Extracted mouse brain via perfusion method, and obtained brain slices on a cryogenic slicer for follow-up immunostaining of specific brain regions.
  • Publication: Phan J, Alhassen L, Argelagos A, et al. Mating and parenting experiences sculpture mood-modulating effects of oxytocin-MCH signaling. Sci Rep. 2020;10(1):13611. Published 2020 Aug 12.

Bioinformatics Projects

  • Residue Counter Script, Python, Developed a reusable Python program to count amino acids or nucleobases in FASTA-formatted sequences. Implemented modular design using custom classes and importable modules to generalize residue counting across different sequence types. Used dictionaries, string handling, and file I/O to tabulate and report residue frequencies, enabling use in broader genomic workflows.

  • Protein Feature Integration Pipeline (Final Project), Python, HMMER, BLAST, TMHMM, Designed and implemented a bioinformatics pipeline to annotate protein sequences using HMMER (conserved domain search), BLAST (homology search), and TMHMM (transmembrane domain prediction). Created individual Python parsers for each tool's output and consolidated results using integrated file output methods.
