Research Project: Isolate and characterize Fe-ADH Thermococcus thioreducens.
Objective: Purification and characterization of Fe-ADH within the T. thioreducens and to see how important iron is for iron-dependent ADH.
• Utilized Bradford assays to determine protein concentration, confirming the presence of the enzyme within the sample.
• Employed activity assays to quantify enzyme activity, establishing the quantity of the enzyme in the sample and its functionality.
• Executed purification techniques to isolate the enzyme
• Investigated and measured enzyme activity under varying cofactor conditions, analyzing and comparing the efficiency of different cofactors in catalyzing reactions.
