  • Thesis Title: Investigating how human behavior such as ethanol consumption affects kaposi sarcoma herpesvirus (kshv) reactivation.
  • Cultured mammalian cell latently infected with KSHV in BSL2 and treated it with ethanol and ethanol metabolites.
  • Used fluorescent microscope for visualization of latent and lytic KSHV infected cells.
  • Determined the percentage of latent and lytic KSHV infected cells using cell counter.
  • Performed BCA & Western blot to determine the protein expression levels of KSHV viral proteins.
  • Conducted qPCR to determine the mRNA expression level of KSHV viral genes.
  • Performed Plate reader assays such as Acetate, ATPase assay and ROS assay on KSHV infected mammalian cells.
  • Carried out genome copy number assay to determine KSHV viral genome copy number.

  • Designed a cold active beta-galactosidase that functions at pasteurization temperature and refrigeration temperatures of 4C for lactose intolerant individuals using molecular cloning techniques.
  • Error Prone PCR/PCR was used to mutate and amplify gene of interest.
  • Isolated the cold active mutant for sequencing to see the sequence change.
  • Identified the phenotype of the cold active mutants and replated them at different temperatures.
  • Characterized the protein by carrying out protein purification and thermo-stability profiles via spectrophotometry.
