I performed AAV9 transduction and tissue culture experiments to administer gene therapy to astrocyte cells derived from ALS patients and skeletal muscle cells from individuals with muscular dystrophy. I reprogrammed skin fibroblast cells into neuro progenitor cells (iNPC) and then differentiated them into astrocyte cells. To confirm the reprogrammed cells' phenotype, I conducted immunostaining using pathological markers.
For an in-depth analysis of lysosomal and mitochondrial changes, along with protein aggregations in treated and untreated astrocyte and skeletal muscle cells, I utilized live cell staining (LysoTracker, MitoTracker), coculture staining, immunostaining, and optimized antibody techniques.
To investigate motor neuron survival and neurotoxicity, I employed coculture with astrocyte cells. Biodistribution studies were conducted through spinal cord cryosectioning with DAB, H&E staining, ELISA, and Laminin/Dystrophin staining.
In the quantification of vector genomes in muscle tissues from muscular dystrophy patients, I utilized ddPCR. Additionally, to measure micro-dystrophin protein expression in treated muscle cells, I followed a protocol involving BSA and Jess. Proteins were extracted from both heart and skeletal muscle for the Pierce BSA protein assay.

Generated a multi-tag positive control protein for expression, purification, mammalian and drosophila cell transfection, Western Blotting. This involved conducting protein purification, western blotting, glycoprotein staining, immuno-precipitation, bacterial transformation, Gel electrophoresis, Microscopy, PCR, and Sanger Sequencing.
Conducted CRISPR gene editing technology to create mutant alleles in Drosophila flight muscles. This research included a reverse genetic screening process, cryosectioning drosophila flight muscles, immunostaining, protein expression and purification for antibody production, fly tests, and pull-down assay to look at the physical interaction of knocked-down proteins with other Z-disc proteins. For the same project, I conducted a CRISPR gene tagging experiment to find the location of the protein of interest.
Operated Transmission Electron Microscopy and Confocal Microscopy for the analysis of Drosophila flight muscle structure for mutation analysis purposes.
Analyzed ESTs, proteins, genomic DNA, and cDNA databases using Blast and ClustalW to identify related proteins and nucleic acids.
Managed hiring and laboratory work orders efficiently.
