ER-Phagy Activation Under α-Synuclein Proteotoxic Stress, Investigated how α-synuclein-induced ER stress activates ER-phagy in S. cerevisiae by quantifying Atg39/Atg40 GFP-tagged receptors using live-cell confocal microscopy. Found a ~2.8× increase in Atg39 and ~1.5× increase in Atg40 during stress, indicating a compensatory autophagy response., Engineered S. cerevisiae strains expressing α-synuclein-HA and GFP-tagged ER-phagy receptors (Atg39-GFP, Atg40-GFP) to model Parkinson's-related proteotoxic stress., Performed live-cell confocal fluorescence microscopy (100x oil objective; GFP/dsRed/BFP channels) and acquired Z-stacks for organelle-level autophagy imaging., Quantified integrated GFP intensity across >100 cells per biological replicate using image-analysis software; performed statistical testing using unpaired t-tests.
