The project focused on the expression and purification of CaVβ3 and CaVβ4 proteins, including a chimeric β4 protein, to explore their interactions with CaVα1 and assess their potential as targets for developing safer chronic pain treatments.

• Executed plasmid DNA transformation, heat shock, and recovery, followed by culture scale-up and protein expression.
• Processed cell pellets via resuspension, French press, and sonication for protein extraction.
• Purified proteins using affinity chromatography, including buffer preparation, sample application, and elution.
• Performed size exclusion chromatography for further protein purification and fraction collection.
• Conducted Western blotting to confirm protein identity, purity, and expression using antibody detection.

The project involved building on previous discoveries to explore mechanisms influencing glucose metabolism and insulin regulation, aiming to develop more effective and lasting therapies for diabetes management.

• Performed PCR genotyping to identify allele types via gel electrophoresis.
• Certified in handling, restraint, injection, and blood collection for mice and rats.
• Administered high-fat diets and glucose/atropine injections to assess metabolism, insulin secretion, and parasympathetic signaling in mice.
• Observed pancreatic resections and analyzed islet morphology through microscopy, linking changes to insulin secretion and glucose regulation.
