Abstract: Assessing the Genetic Diversity of Endangered Lady's Slipper Orchids Local to New England > Showy lady's slippers orchids (Cypripedium reginae) are a critically endangered species in the New England area. Lady's slippers are generally known to grow in small isolated populations and have particular habitat preferences. Additionally, lady's slippers reproduce both through sexual and asexual methods. The aim of the experiment is to see the genetic diversity of showy lady slipper's populations in New England, specifically in Eshqua Bog Natural Area in Vermont. Low genetic diversity within a species can lead to the extinction of the species in a changing and industrialized world. To determine genetic diversity, 39 different primers were used on multiple samples of DNA within this showy lady's slipper population to target specific microsatellite regions with the use of PCR. Since some of these primers were new to the lab, an additional experiment was performed to determine the annealing temperatures for these primers. After sequencing the samples made during this experiment, the differences and similarities between individuals within this population were evaluated to determine their genetic diversity. This research will contribute to the future goal of strengthening populations of showy lady's slippers in the New England area by increasing the genetic diversity within them.

Abstract: Identification of Cannabinoids in Commercial CBD Oils Using High-Performance Liquid Chromatography (HPLC) > The increasing popularity of cannabidiol (CBD) products has raised concerns regarding their safety, accuracy of labeling, and cannabinoid composition. This study utilizes high-performance liquid chromatography (HPLC) to measure the concentrations of CBD, tetrahydrocannabinol (THC), and cannabinol (CBN) in various commercial CBD oils. Six different brands of CBD oils were analyzed using an HPLC system with a reverse-phase chromatography method to separate and quantify cannabinoids based on polarity differences. A calibration curve was established using standard cannabinoid solutions to ensure accurate quantification. The results indicate variability in CBD concentration compared to labeled values, with some brands displaying discrepancies between reported and measured amounts. Notably, Dr. Formulated and Foster Botanicals exhibited higher-than-labeled CBD concentrations. Future research will aim to expand the study to detect additional cannabinoids beyond CBD, THC, and CBN and investigate their pharmacological effects. This study underscores the need for rigorous quality control in the commercial CBD market to ensure consumer safety and accurate product labeling.

Abstract: The Effects of Microplastics on the Digestive Tract, Fertility, and the Body of C. elegans > Microplastics are an emerging environmental concern due to their potential effects on biological organisms. This study investigated the impact of fluorescent microplastics on the fertility, digestive tract, and overall body of the nematode Caenorhabditis elegans (C. elegans). C. elegans were exposed to microplastics at concentrations of 3.5 mg/mL, 1.75 mg/mL, and a 0 mg/mL control group. Fertility was assessed by counting the number of eggs laid and the percentage of unhatched eggs. The digestive tract and body retention of microplastics were examined using fluorescence imaging. Our results indicated that microplastics remained inside the digestive tract of C. elegans for up to eight days post-exposure, with fluorescence intensity significantly higher than controls (p C. elegans showing difficulties in egg-laying. While T-tests revealed no significant differences in fertility metrics between experimental and control groups except for the highest concentration on day 2 (p = 0.05), the accumulation of microplastics within the body suggests potential long-term biological effects. Further research is needed to confirm these findings and explore potential implications for other organisms, including humans.
