We are investigating the carcinogen-induced lung tumorigenesis. Hexavalent chromium (Cr(VI)) reduces the protein level of the DNA polymerase δ subunit p12 in lung epithelial and lung cancer cells by promoting its degradation through the RNF8- and CRL4Cdt2-dependent ubiquitin–proteasome pathway. This loss of p12 shifts Polδ from its tetrameric Polδ4 form to Polδ3, leading to increased DNA damage, stronger activation of the DNA damage response, and reduced cell proliferation and survival after Cr(VI) exposure. These findings show that Polδ4 helps maintain genome stability during Cr(VI)-induced stress and that p12 loss may contribute to chromium-related lung carcinogenesis. We are also studying BaP, NNK and Urethane-induced lung carcinogenesis.

Mitochondrial dysfunction delays mitochondrial protein import, causing accumulation of non-imported mitochondrial proteins in the cytosol and challenging proteostasis. Cells respond by increasing proteasome activity and molecular chaperones in yeast and C. elegans. In humans, Immunoproteosome is activated upon mitochondrial dysfunction. Still, several consequence mechanisms were not completely understood between mitochondria and UPS.
Performed biological experiments to identify the molecular mechanisms underlying mitochondria and the Ubiquitin Proteasome System.
