Work History

Associate Research Scientist, Drug Substance Development / Process Development

OCUGEN OPCO, INC.
Malvern, PA
11.2022 - Current
  • Support the cell therapy manufacturing process, production of recombinant protein, and AAV expression in mammalian host systems.
  • Support the upstream process (USP) and downstream process (DSP) scale-up to support early and late-stage projects.
  • Process Development of the pipeline projects for pre-clinical and clinical applications.
  • Currently focused on a project called NeoCart. It is a type of regenerative medicine / tissue engineering therapy for knee cartilage repair.

Protein Drug Development Formulation Scientist

EUROFINS LANCASTER LABORATORIES PSS
Malvern, PA
01.2017 - 11.2022
  • Performed experimental studies related to formulation, process development and testing of therapeutic proteins (monoclonal antibodies), virus-like substances and gene therapy related molecules.
  • Tests performed included MFI (micro-flow digital imaging) using Protein Simple MFI 5200 5000 Series, HIAC (High Accuracy Products; sub-visible liquid particle counting) using Beckman Hach HIAC 9703+, turbidity, pH, osmolality and color determination.
  • Proficient in Biovia Electronic Laboratory Notebook, GraphPad and Vector NTI.

Vector Production Manager / Phillip Johnson Laboratory, Department of Infectious Diseases

CHILDREN’S HOSPITAL OF PHILADELPHIA RESEARCH INSTITUTE
02.2007 - 12.2016
  • Produced and purified recombinant AAV viral vectors under GMP conditions by FPLC (using the ÄKTA Explorer Model 100A system powered by UNICORN software in conjunction with Cytiva HiTrap 1ml Sepharose cation exchange columns) for use as experimental vaccines.
  • Titered purified vectors using the ABI FAST Real-Time Quantitative PCR System.
  • Cloned and purified vector plasmids used for calcium chloride transfection.
  • Maintained cell lines (mostly HEK 293) used for vector production.
  • Performed in vivo studies in rabbits to evaluate the safety and efficacy of vectors as potential vaccines that comply with FDA Investigational New Drug submission guidelines.

Research Specialist / Department of Anesthesia

UNIVERSITY OF PENNSYLVANIA PERELMAN SCHOOL OF MEDICINE
03.2002 - 01.2007
  • Characterized the proteome of a novel glutamate receptor-associated transcription unit, GRINL-1A, by (1) antibody inhibition of NMDA cytotoxicity in cultured cortical and hippocampal neurons, (2) Western blots and (3) immunofluorescent staining (http://www.ncbi.nlm.nih.gov/pubmed/15233991).
  • Used bacterial cloning to express and purify a peptide containing the putative anesthetic binding site of the nicotinic acetylcholine receptor alpha M2 subunit. This peptide was then used in NMR-based structural biology studies to refine the understanding of the ligand-receptor interactions of anesthetics.

Research Specialist / Department of Neurology, Neuro-oncology Division

UNIVERSITY OF PENNSYLVANIA PERELMAN SCHOOL OF MEDICINE
01.2005 - 01.2006
  • Performed immunoblots of patient sera and cerebrospinal fluid against proteins from cultured neuronal cells that express antigens associated with paraneoplastic neurologic syndromes (PNS).
  • See: http://onlinelibrary.wiley.com/doi/10.1111/j.1750-639.1999.tb00226.x/abstract.
  • Performed immunohistochemistry of frozen rat brain sections using sera from patients with PNS.
  • Screened cerebellar and brainstem cDNA libraries for PNS-associated antigens using phage display. The resulting data was then used to determine distinct regions of the brain involved in the pathology of PNS.
  • Performed molecular cloning, PCR, immunofluorescent staining, immunoprecipitation, and Western blots.

Research Specialist / Institute for Human Gene Therapy

UNIVERSITY OF PENNSYLVANIA PERELMAN SCHOOL OF MEDICINE
01.2002 - 01.2005
  • Produced and purified recombinant adenovirus and AAV vectors in compliance with GLP/GMP standards.
  • Cloned plasmids for production of viral vectors by transfection of mammalian cell lines.
  • Analyzed transgene expression by Western / Southern blot, ELISA, FACS and PCR.
  • Performed immunological assays to detect pre-existing neutralizing antibodies to AAV vectors.
  • Performed quality control / quality assurance assays and documentation for titer of infectious viral particles, presence of replication competent adenovirus and endotoxin. These assays were necessary to ensure the purity and safety requirements of vectors before proceeding to in vivo studies.
  • Performed PCR genotyping and maintained breeding records for transgenic mice which were used to study the role of beta-defensin 1 in pulmonary innate immunity (http://www.ncbi.nlm.nih.gov/pmc/articles/PMC127957/).
