Work History

Undergraduate research assistant

University of Arizona - Betül Kaçar
Tucson
09.2018 - 08.2021
  • Preformed Lambda red recombineering removed the native elongation factor gene (tufA) and replaced it with a foreign elongation factor
  • Followed by a long term evolution study performed on 6 mutant strains of E
  • Coli, tracking fitness changes and mutations over time
  • In order to observe differences in evolution due to the replacement of an essential gene with a foreign variant
  • Created algorithms during the initial phases of building a phylogenetic tree of Initiation factor (IF2) which would aid in simplifying steps towards organizing and editing data files
  • Computational Analysis of essential proteins involved in the translation machinery using software to generate highly probable phylogenetic trees followed by ancestral sequence realignment to assess the molecular identity of essential genes in correspondence to how they might have looked on early earth
  • Genetically engineered ancestral reconstructed versions of Initiation factor (IF2) involved In translation machinery into modern day Escherichia coli homolog
  • Similarly to Escherichia coli I worked with another colleague in reengineering Cyanobacteria with the ancestral reconstructed form of the large subunit in Rubisco, an essential enzyme responsible for photosynthetic and chemoautotrophic carbon fixation and oxygen metabolism
