Metropolitan State University, Bacteriophages were isolated from environmental filtrates and screened against 12 BSL1 bacteria using a spot assay. Raw sewage showed the most promising results with forming plaques on the E. coli K12 plate. Other strains of E. coli (EC-B and EC-C) were cultured and used to further specify the host range of the phage. After inoculation and incubation, the cultures were serial diluted (10^1-10^10), and all sequential dilutions were plated with their corresponding E. coli strain. EC K12 showed the most substantial results on dilutions 101 and 102. Plaques were removed from these plates and incubated in cultures of EC K12. The most evident result was in culture (#3), which indicated high levels of lysis via complete clearing of the culture’s turbidity. Samples from culture #3 were used for a spot assay against the 12 BSL1 bacteria. The only plate with plaque formation was EC k12., This Phage is currently being used in the lab portion of the Microbiology course at Metropolitan State University to type E. coli. The phage has not been sequenced and is currently named Funka3.
