MUTAGENESIS OF THREE PUTATIVE CHROMIUM REDUCTASE GENES IN SERRATIA MARCESCENS.

• Prepared media and cultured S.marcescens isolates.
• Introduced a plasmid derived from Escherichia coli into the cultured S. marcescens using transformation and conjugation assays.
• Purified the kanR gene from another plasmid and amplified it using polymerase chain reaction (PCR).
• Conducted electroporation of S. marcescens with the kanR gene to facilitate homologous recombination.
• Performed PCR and agarose gel electrophoresis to confirm the successful deletion of the target genes.

IDENTIFICATION AND CHARACTERIZATION OF SOIL BACTERIA ISOLATE, RESISTANT TO ESKAPE PATHOGENS

• Collected soil bacteria isolates and cultured using serial dilution.
• Sub-cultured the bacteria by patching and streaking to obtain new individual colonies whilst conducting antibiotic susceptibility testing using the disk diffusion assay.
• Sub-cultured successful isolates that exhibited zones of inhibition against the ESKAPE pathogens.
• Amplified the 16S ribosomal RNA gene of successful bacterial isolates using polymerase chain reaction and gel electrophoresis.
• Characterized the bacteria after DNA sequencing using NCBI BLAST.

COMPARISON OF CYANIDE CONTENT IN THREE TYPES OF GARRI SOLD WITHIN OWERRI NORTH, NIGERIA.

• Collected garri samples from four local markets using a multi-stage sampling technique.
• Analyzed the samples for cyanide content in triplicate using a multi-parameter method.
• Developed research hypotheses and analyzed the sample data for significant differences in cyanide content using SPSS ANOVA test (Analysis of Variance).
