  • Lead participant in the investigation of bacterial persistence mechanisms of the pathogen Porphyromonas gingivalis (PG W83)
  • Conducted bacterial growth curve and survival curve assays in context of sublethal exposure to tetracycline and metronidazole antibiotics using both optical density (OD550) and viable colony formation units (CFU) approaches
  • Prepared lyophilized concentrated culture filtrate (CCF), based on the hypothesis that bacteria secrete factors that regulate persister cell development in bacterial populations
  • Developed protocols and optimized assays to quantify changes in the percentage of persister cells amongst PG W83 cultures, with or without CCF treatment, followed by an overnight exposure to sublethal dose of antibiotics of samples collected at select time-points during growth
  • Gained experience in custom primer design for RT-qPCR analysis of PG RNA targets putatively involved in bacterial persistence based on published studies and using DNA, RNA, and protein databases and bioinformatic online tools
  • The selected PG targets were identified by sequence homology to genes identified in other bacteria (e.g., E
