  • Research involved RNAi reagent development in the mouse macrophage RAW264.7 cell line (AfCS model cell type), RNAi optimization included: siRNA transfections, shRNA construction and testing (transient transfections), and production of shRNA expressing lentiviral and retroviral cell lines
  • Tested large batches of siRNAs and shRNAs (single and in tandem) targeted to various genes involved in G-protein signaling pathways
  • Performed extensive cloning, vector construction using Gateway Technology from Invitrogen, retroviral and lentiviral production and purification, maintained cell lines (RAW 264.7, Phoenix cells and HEK293T), harvested and cultured primary mouse macrophages (from bone marrow), transfected and infected various cell lines, RNA and protein isolation, qRT-PCR as well as traditional PCR, Western blots, Southern blots, immunprecipitations, phagocytosis of macropages derived from bone marrow and site-directed mutagenesis
  • Monitored changes in G-protein coupled receptor desensitization due to various gene knockdowns
  • Maintained transgenic rodent model, included setting up breeding pairs and general management of small animal colony (PCR-based genotyping)
