Work History

Associate Scientist II

AbbVie
Worcester, MA
04.2019 - Current

•Designed and executed experiments utilizing laboratory techniques to advance the development of Antibody Drug Conjugates for the treatment of immune-mediated diseases and HIV.

• Established 3D mammalian cell culture methodology to evaluate small molecules and antibody drug conjugates, with the aim of improving predictions of efficacy in vivo.

• Implemented and trained others in the use of the ImageXpress micro confocal high content imaging system and developed plate-based immunofluorescence techniques for 2D and 3D cell cultures to study binding, trafficking, and mechanism of action of therapeutic candidates, as well as other antibodies and small molecules.

• Created experiment specific custom analysis modules to effectively quantify data from high content imaging studies using the MetaXpress software.

• Pioneered an innovative and cost-effective in-vitro 3D spheroid tumor penetration model to assess biologics of various target affinities and sizes over multiple timepoints via high content cellular imaging.

• Designed intricate analysis methodology for comprehensive quantification of biologic penetration into 3D tumor spheroids.

•Presented advancements and outcomes from tumor penetration model development at yearly internal meetings.

• Cultivated and maintained a diverse range of mammalian cell lines, including primary cells, reporter lines, and isolated Human PBMCs.

• Utilized multiple transfection methods, including reverse transfection, nucleofection, and lentiviral vectors in mammalian cells.

• Verified protein expression and degradation through Western Blot analysis

• Validated efficacy of small molecules and antibody drug conjugates through ELISA, cell proliferation, and reporter assays.

• Extracted RNA and DNA from mammalian cells and developed TaqMan (q-PCR) biomarker assays to assess target modulation and expression.

• Conducted secondary saporin toxin assays to evaluate antibody internalization in mammalian cells.

• Designed and executed binding and internalization experiments for multicolor flow cytometry and real-time cell imaging with Amnis Imagestream.

• Utilized Tecan microfluidics system for the titration and addition of small molecules to plated cells.

• Formulated media, chemicals, and managed an electronic lab notebook for efficient record-keeping and data management

Research Associate

• Utilized laboratory techniques to support the development of a novel exosome-based drug delivery system.

• Isolated proteins using Tangential Flow Filtration, Density Gradient Centrifugation, Size Exclusion Chromatography, and SDS-PAGE

• Verified protein expression through standard and chemiluminescent Western Blot analysis and assessed protein concentration using various quantitative assays such as BCA, micro BCA, and Qubit Assays

• Managed and maintained a diverse range of mammalian cell lines, including reporter lines such as A549-LUC and Cre-Lox (HEK 293)

• Conducted MTT and cellular uptake assays, as well as uptake experiments in seedlings

• Evaluated the impact of drug delivery candidates on inflammatory cytokine production through Macrophage ELISA

• Formulated media and chemicals to support a range of experiments and effectively managed laboratory organization and stocking

• Management of an Electronic Lab Notebook

Student Researcher

Tufts Veterinary School
Grafton, MA
09.2017 - 08.2018

• Contributed to a laboratory project under the supervision of Dr. Sam Telford III to standardize an ELISA for Deer Tick Virus

• Purified monoclonal antibody HuMab 5C12 for the detection of shiga toxin 2 in sera, and characterized purity and presence using ELISA, SDS-PAGE, and Western Blot

• Assessed the efficacy of HuMab 5C12 in protection against STEC in Vitro and in Vivo

• Conducted experiments to propagate Cryptosporidium sp. in an immunosuppressed mouse model

• collected and analyzed fecal samples and gastrointestinal tissues, and performed histopathological and electron microscopy examination

• Isolated genomic and specific DNA from the human parasite S. mansoni, amplified genes using rt-qPCR and qPCR, cloned genes using competent E. coli expressing recombinant protein, and characterized basic biochemical properties

• Conducted experiments to analyze the kinetics of converting or dephosphorylating substrate pNpP into the product p-nitrophenol (pNp) using S. mansoni recombinant alkaline phosphatase
