Project title: Investigation into the lesion bypass activity of human DNA polymerase kappa following a mutation of the residue D109N in the active site. (Mentor: Dr Samer Lone)
Project involved the mutation polymerase kappa, a member of the Y family polymerases, to test the lesion bypass activity.
Site directed mutagenesis and a series of protein purification steps were performed followed by DNA polymerase assay.
The tests performed were to determine if a mutation occurred in the active site of pol K where the binding site for replication is located. The chosen mutation was theorized to inhibit or eliminate all replication activity. Without further testing the mutation inhibited all replication activity.
