- Teaching Assistant in Molecular Microbiology Course for the cloning laboratory where the E. Coli bacteria takes up the recombinant pJET 1.2 plasmid with the GAPDH gene inserted into the eco47IR sequence.
- Instructed students on size exclusion chromatography to purify PCR product.
- Blunt end ligation was then instructed to the class to join the vector DNA and insert the DNA.
- Then last portion of the the laboratory was the transformation process, introducing the plasmid into living bacterial cells so replication can occur. The cells were placed in a CaCl ice bath and then heat shock was utilized to create competent cells.
- Cells were incubated on LB amp IPTG plates. E. Coli containing the recombinant plasmid grew on most plates proving the pJET 1.2 contains the eco 47IR gene.
