Research on the role of Cadherin-11(CDH11) in mediation of triple negative breast cancer metastasis, 08/2019-08/2021

  • Tools and techniques : Cell culture, Western blot, Silver staining, TMA analysis, RT-PCR, Cloning, Bradford assay, Top-Flash assay, TCGA data analysis, Immunohistochemistry, Chip-seq, CRISPR/Cas9, Lenti-virus production, Cellular compartment assay, Confocal Microscopy, Cell proliferation assay, migration assay and invasion assay, etc.
  • Data analysis based on Histology staining of large-scale of Tumor micro-array (TMA) via Qupath to detect positive signaling as well as subcellular location in order to establish the correlation between patients and targeting proteins expression.
  • Generating knocking out cell lines via CRISPR/Cas9 and comprehensively characterized cell lines properties via RNA-seq, Western Blot and tumor related functional assays.
  • Introduced a Wnt-pathway reporter in the cell lines and monitor the Wnt-pathway activities in a real-time manner via TOP-Flash Assay. This system was employed as a platform to quantitatively select drugs and antibodies that targeting tumorigenesis related Wnt-pathway.
  • Utilized a high resolution confocal microscope to generate 3-D model of a single cell to depict the signaling location.

Research on the role of CDH11 in mediation of cell growth and promote wound healing in vivio, 12/2016-08/2019

  • Tool and techniques: KO mice, in vivo cutaneous wound healing, Hisotology, Western blot, Immunostaining, Immunoprecipitation, Viral production, Cloning, SiRNA, RT-PCR, CDH11-Fc coated surface, etc.
  • KO mice and WT control were employed to study the deficiency effect of CDH11 in vivo. Multiple tissues were isolated for immunostaining in order to evaluate targeting protein expression.
  • Molecular biology technique was utilized to develop multiple different functional domains depletion CDH11 proteins
  • Kinetics experiments was performed to capture the signaling cascade and biochemically blocking various signaling pathways to illuminate the signaling propagation.

Research on the role of CDH11 in regulation of Extracellular Matrix (ECM) synthesis and tissue mechanical properties, 01/2014-12/2016

  • Tool and techniques : KO mice, Instron, Tissue Organ Bath system, Tissue isolation, Cell isolation, Histology, Immunostaining, Cloning, Western Blot, SiRNA, etc.
  • Multiple organs and tissues were isolated from KO mice and WT control to determine the mechanical force generation in vitro by tissue organ bath system.
  • Tissue Mechanical properties were measured and quantitatively reported by Instron.
  • ECM contents in different organs were determined by both immunostaining and biochemistry.
  • CDH11-Fc coated surface was utilized to mimic the naturally CDH11 homotypical interaction and detect the early event signaling.
