Academic Conference

  • I attended the 2nd International Research Conference on Neurodegenerative Diseases (IRCND 2022) from July 17 to 20, 2022, at the Scott Conference Center in Omaha, Nebraska. A poster presented by a lab member was awarded the Young Investigator Award for its exceptional presentation.
    The winning team consisted of Devanshi Shukla, Ju Gao, Luwen Wang, Xiaojia Ren, Zhongbin Wang, Justin R. Dunn, Ariele Peters, and Xinglong Wang. The research focused on the "Correlation Between SARS-CoV-2 Structural Proteins and Tau," which was presented on July 19, 2022, as part of the IRCND Meeting.

The feeding and management protocols for the laboratory mouse facility have been meticulously crafted through a comprehensive review of pertinent documentation and careful consideration of the mouse cage placements, ultimately leading to a significant reduction in the valuable time spent by the facility's personnel. This streamlined management approach has been consistently applied across the laboratory.
The implementation of an adjustable spacing pipette significantly streamlined the process of genotype identification, lessening the burden on laboratory personnel and elevating overall operational efficiency. This innovative method was universally adopted across the laboratory.
Thousands of mice were obtained to manage and monitor the disorder movements in brain tumor mouse models, specifically those involving Nf1-related and MADM mutant mice. Furthermore, daily monitoring of mice predisposed to brain tumors was consistently conducted, accompanied by sampling and subsequent experimentation, which encompassed dissection and immunostaining analysis of various locations within the brain tumors of these mice.

The construction and purification of insoluble proteins, specifically TDP43 LCD protein, which is associated with Alzheimer's disease, has been undertaken. We successfully conducted an assay to investigate the liquid-liquid phase separation (LLPS) of TDP43 LCD protein in vitro, following the addition of varying concentrations of NaCl. This endeavor validated the mechanism behind the common disease phenotype arising from TDP43 protein droplets formed through LLPS.

The mitochondria extracted from the cells of TDP43 knockout mice suffering from brain disease was carried out using an O2K equipment for mitochondrial function assay. The O2K technology will be instrumental in demonstrating indicators of abnormal oxidative phosphorylation in brain disease, specifically regarding the respiration rates of complexes I, II, and IV.

The molecular mechanism by which morphine-induced RAB31 mediates the release of extracellular vesicles (EVs) in astrocytes derived from C8D1A mouse cells underscores the crucial role that RAB31 plays in the release of EVs.

The specific function of the mouse testis-specific gene TBC1d21 is to regulate spermatogenesis. I have constructed 30 eukaryotic and 30 prokaryotic expression vectors, which can be utilized for conducting cell transfection experiments and analyzing the enzyme activity of the target protein.
Preparation of point mutation mice involves the careful selection of amino acid sites for the introduction of mutations, the meticulous design of guide RNAs (gRNAs) and primers specifically tailored for identifying these mutations. Subsequently, unique primers are devised to ascertain the genotype of F0 mice harboring the point mutation. Following this, by comparing the sequencing outcomes of the offspring, the genotype of the mutated mice is definitively determined.

China National 863 project-Establishment and Application of a Transgenic Platform for Chickens
I was the pioneer in discovering that the endotoxin level, specifically 0.05EU/μg versus 0.1EU/μg of plasmid DNA, plays a pivotal role in animal experiments in vivo. My findings revealed that this endotoxin level can adversely affect transgenic efficiency in transgenic animals. To address this challenge, I identified a kit known as "NucleoBond® XtraEF kits, REF. 740424.10/.50," which consistently yields purified DNA that is virtually free of endotoxins( Plasmid Kits, which guarantee endotoxin levels of less than 0.1EU/μg plasmid DNA. This nuanced understanding highlights the need for meticulous control over endotoxin levels in transgenic platforms.
In addition, the published paper has been reproduced by renowned domestic research institutions, including the HARBIN VETERINARY RESEARCH INSTITUTE, the Chinese Academy of Agricultural Sciences (CAAS).
