  • Designed & developed high parameter immunophenotype panels on BD FACSlyric for characterization of starting material and DP for (Chimeric Antigen Receptor) CARNK and Gamma delta cell therapies platform.
  • Optimized BD Lyric by performing voltage titration for immunophenotype assay development.
  • Prequalified immunophenotype assay to establish the robustness, precision and specificity for DP release and collaborated with cross functional teams for tech transfer and routine testing.
  • Optimized and validated the immunophenotype assays and performed fit for purpose assay validatation.
  • Designed Flourescne minus multiple (FMM) in place of FMO controls for feasabiility of tech transfer.
  • SME for flow cytometry in the AD department.
  • Executed infectious titer assays to assess transduction efficiency and to quantitate CAR expression.
  • Executed cytotoxicity assays to assess the potency of CARNK DP.
  • Conducted proliferation assays aimed at assessing the proliferation rate of NK cells.
  • Executed surface vs intracellular staining to compare the CAR expression in DP vs UTD.
  • SME for critical reagent testing which included in house generated anti CAR antibody clones to identify best clone for CAR detection.

  • Performed cell culture to maintain cell lines utilized in cytotoxicity studies. Banked cells like raji, K562, daudi cells and made Master Cell Bank (MCB) and Working Cell Bank (WCB) using Cryomed freezer.
  • SME for stability testing for determining the stability of DP overtime by testing at different timepoints for its appearance, extraction volume, cell count and viability, immunophenotype.
