Project 1 – Role of molecular chaperones in Alzheimer’s disease
- Designed the project and performed molecular biology scientific research using ELISA, Immunofluorescence microscopy, Western blot, BN-PAGE analysis, Immunoprecipitation, siRNA, shRNA, PCR, and Extraction of RNA from cells.
- Used various small molecular inhibitors to discover the drug for Alzheimer's disease.
- Collaborated with colleagues in other departments to develop a drug for Alzheimer's disease.
- Submitted in the Journal of Biological Chemistry.
Project 2- Familial Alzheimer's disease mutations induced structure, dynamics, and functional alteration of the gamma-secretase complex
- Performed a series of molecular biological experiments to investigate functional characterization of gamma-secretase complex.
- Developed HEK293T cells stably overexpressing APP-C99 and its mutant to investigate enzyme-substrate complex formation by Immunoprecipitation.
- Developed the technique to analyse the enzyme-substrate complex by using BN-PAGE.
- Participated with a research team to develop mouse models for projects.
- Prepared IP samples for proteomics studies and collaborated with the University's mass spectrometry lab to conduct the proteomics research.
- Used neurons from NPCs derived from iPSCs to explore enzyme-substrate complex formation in mutations linked to Familial Alzheimer's disease.
- Contributed to publication in Cell Reports journal by writing manuscripts, preparing figures, and editing text.
Project 3- Effect of Familial Alzheimer's disease mutations on mitochondrial function
- Used Seahorse XF mito stress test to measure oxygen consumption rate in HEK293T cells carrying FAD-linked mutants.
- Isolated the mitochondria from the cells.
- Provided technical guidance to undergraduate students working with me on the projects.

Project- Effect of high temperature on amyloid beta production
- Discovered that high temperatures can increase the production of amyloid beta, a protein associated with Alzheimer's disease, through the activity of heat shock protein 90 (Hsp90). This suggests that high temperature may be a novel risk factor for Alzheimer's disease.
- Performed molecular biology scientific research using methods including ELISA, Western blot, BN-PAGE Immunostaining, PCR, Immunoprecipitation, and shRNA.
- Developed the technique for Immunoprecipitation by using 1% digitonin as a detergent to observe the interaction between gamma-secretase components and other proteins.
- Designed BN-PAGE protocol for the lab.
- Developed shRNA Hsp90 knockdown stable HEK293T cell line.
