  • Development of novel clinically relevant models of oral/head and neck cancers such as primary monolayer, spheroid and organoid (in vitro) and PDX, syngeneic and humanized mouse models
  • Translational investigation of disease mechanisms in head and neck preclinical models by investigating essential cancer pathways that related to DNA repair, cell cycle, EGFR signaling that related to PI3K/ERK/AKT/STAT3 pathways.
  • High-throughput drug screenings in collaborations with drug screening core facilities by screening FDA approved drug library with 1506 compounds in organoid models.
  • Non-invasive imaging such as magnetic resonance imaging monitor longitudinally of tumor growth in vivo and bioluminescent imaging such as luciferin to detect longitudinally of tumor growth and using firefly luciferase (Fluc) and luminol in tumor of in vivo.
  • Developed organoid -based assays to identify the mechanistic role of various targets in cancer using small molecule that inhibit the target expression and determine the therapeutic response to standard of care treatment.
  • Identify novel therapeutic agents using drug repurposing approach with and without standard of care regimens in preclinical models.
  • Developed organoid-immune cell (CD3 and CD8 T cell) co-culture assay to evaluate the immunomodulatory efficacy of lead compound using ELISA and validate using syngeneic or humanized head and neck cancer model through flow cytometry.
  • Utilized flow cytometry for immune-phenotyping (10 fluorochrome colour panel) to measure T cell functionality such as IFNG and Granzyme B.
  • Performed several immunoassays to analyze the cytokine levels in cell cultures supernatant in response to various treatments.
  • Analyzed post-transcriptional and translational effect after treatment by measuring gene expression via qPCR and protein levels by western blot.
  • Develop stable knock out cell lines using either plasmid transfection or lentivirus transduction.
  • Evaluated and implemented CyTOF and Nanostring spatial transcriptomic sequencing.
  • Resulted in four publications through collaboration with other scientists, which relate to the translational effects of using small molecule therapeutic agents in cancer preclinical models.
