  • I worked with CRISPR-Cas9 genetic manipulation, including molecular cloning, lentiviral transduction, and culturing various cell lines such as HepG2, HEK293, and U2OS. My primary responsibilities involved performing multiple gene knockouts (KO) using lentiviral transduction, conducting DNA extraction, PCR amplification, and PCR product purification, as well as analyzing DNA sequences and chromatograms using SnapGene. Two of the main projects I contributed to included establishing a TASOR KO rtTA BFP U2OS cell lines for future experiments, and cloning a dPAM-dCas9-P2A-mCherry plasmid for future lentivirus production and infection.

CD133: An Immunotherapeutic Target Against Hepatocellular Carcinoma

  • This independent project aimed to increase the expression of CD133, a cell-surface glycoprotein that has the potential to become a molecular biomarker for hepatocellular carcinoma, and therefore, a more viable target for immunotherapeutic options such as CAR T-cell therapy. HepG2 cells were stimulated with IL-6 in order to increase the expression of CD133. This cytokine was chosen as the stimulant due to its role in the JAK-STAT pathway. This project was an award winner of the 2025 MCPHS Center for Research and Discovery Conference.

Testing the Anti-Inflammatory Property of Chalcones

  • This independent project, funded by an MCPHS mini grant, aimed to develop anti-inflammatory chalcone derivatives that reduce cytokine production in stimulated cell cultures. Chalcones were synthesized via aldol condensation reactions using benzaldehydes (4-hydroxy-3-nitrobenzaldehyde and 4-(1H-imidazol-1-yl)benzaldehyde) and various acetophenones. The derivatives were tested in intestinal epithelial cells (HT-29) stimulated with LPS to induce cytokine production (IL8).
